چکیده

There is an increasing demand for versatile and ubiquitous Cytochrome P450 (CYP) biocatalysts for biotechnology, medicine, and bioremediation. In the last decade there has been an increase in realization of the power of CYP biocatalysts for detoxification of soil and water contaminants using transgenic plants. However, the major limitations of mammalian CYP enzymes are that they require CYP reductase (CPR) for their activity, and they show relatively low activity, stability, and expression. On the other hand, bacterial CYP enzymes show limited substrate diversity and usually do not metabolize herbicides and industrial contaminants. Therefore, there has been a considerable interest for biotechnological industries and the scientific community to design CYP enzymes to improve their catalytic efficiency, stability, expression, substrate diversity, and the suitability of P450-CPR fusion enzymes. Engineered CYP enzymes have potential for transgenic plants-mediated phytoremediation of herbicides and environmental contaminants. In this review we discuss: 1) the role of CYP enzymes in phytoremediation using transgenic plants, 2) problems associated with wild-type CYP enzymes in phytoremediation, and 3) examples of engineered CYP enzymes and their potential role in transgenic plant-mediated phytoremediation.

Challenges and Opportunities of Cytochrome P450-Mediated Phytoremediation.

Although environmental pollution was first realized to be a threat to public health in the latter part of the 20th century, events in the 21st century have served to increase the public awareness of the impacts of environmental pollution. Several communities, including the scientific community initiated dialogues to help protect the planet from environmental pollution. Further, scientific commu...

Enhanced metabolism of halogenated hydrocarbons in transgenic plants containing mammalian cytochrome P450 2E1.

Chlorinated solvents, especially trichloroethylene (TCE), are the most widespread groundwater contaminants in the United States. Existing methods of pumping and treating are expensive and laborious. Phytoremediation, the use of plants for remediation of soil and groundwater pollution, is less expensive and has low maintenance; however, it requires large land areas and there are a limited number...

Transgenic plants for enhanced phytoremediation of toxic explosives.

Phytoremediation of organic pollutants, such as explosives, is often a slow and incomplete process, potentially leading to the accumulation of toxic metabolites that can be further introduced into the food chain. During the past decade, plants have been genetically modified to overcome the inherent limitations of plant detoxification capabilities, following a strategy similar to the development...

Transgenic Rice and Potato Plants Expressing Human Cytochrome P450s Show Cross-tolerance to Herbicides by Detoxifying Them

Transgenic plants expressing various species of the detoxifying enzyme, cytochrome P450 monooxigenase from mammals, were bred by Agrobacterium-mediated transformation. The transgenic plants metabolized exogenous chemicals, including herbicides which they were able to tolerate. Because the enzymes had broad substrate specificity, the transgenic plants showed crosstolerance to several herbicides,...
